First (and more common): It is a way of asking you to do something that you do not have to do, are not obligated to do. For example, in a supermarket, a short woman might say, "Excuse me, Sir, could you do me a favor and hand me that package on the top shelf? I cannot reach it." Usually, you use it for a situation that is more serious than the supermarket - "Kyrene_Hua, I know you are busy, but could you do me a favor and call our boss and tell him/her that we need an answer now for our project."

Second (lesson common - sarcasm): If someone is angry with you or frustrated about a situation with you. For example if you said, "Hey, WiseRichard, would you do me a favor and stop telling those bad jokes of yours?" (I had better do that if I want you to call our boss for me).

In a conversation, the tone of voice will tell you which one it is (polite or sarcasm). In writing, the context will tell you which one it is.
"Hey, WiseRichard, would you do me a favor and stop telling those bad jokes of yours?"
"Well, I like them."
"Have you noticed that nobody laughs at them. Even the cats leaves the room." Forget polite in this context.
