A: "Ain't" es una contracción no estándar que se usa en unos dialectos norteamericanos y británicos. En muchos de esos dialectos, puede sustituye por "am not", "aren't" y "isn't".

I ain't going to do it. = I'm not going to do it.
He ain't a teacher. = He isn't a teacher.
We ain't going to school. = We aren't going to school.
They ain't our friends. = They aren't our friends.
Ain't it the truth? = Isn't it the truth?

También puede sustituye por "haven't" y "hasn't", especialmente en inglés británico:

I ain't been to the store. = I haven't been to the store.
He ain't eaten. = He hasn't eaten.

En inglés vernáculo afroamericano, puede sustituye por "didn't":

I ain't know what he said. = I didn't know what he said.

A: Neither. You say "her mother is". It is a possessive pronoun and needs no " 's ". Similarly "His mother is" when talking about a man's mother, and "it's mother " (note no apostrophe) for an animal's mother.
