Your meaning is not clear. Do you want “1 day off every week”? Or “after one week, you want one day off”?

I need one day off in a week - means sometime next week you will need a day off to do something, doesnt matter what day it happens to be
Can I have one day off in a week - again says next week you will need one day off but it doesnt matter what day it is. In this one you are asking for permission by asking if you can. In the pervious one you made a statement.
I need a day off next week - also says you need a day off but sounds a little more natural and tells your boss that you absolutely need the day off. You are telling him, not asking.
