La primera frase debería decir "Does my neighbor play with me?"= "¿Mi vecino juega conmigo?" Significa algo que ocurre con regularidad.
La segunda dice "¿Está jugando mi vecino conmigo?". Significa algo que actualmente ocurre.
La diferencia en inglés entre el presente y el progresivo importa más que en español. Puedes decir "Como tacos" en español si estás comiendo tacos ahora mismo pero en inglés hay que decir "I'm eating tacos" porque "I eat tacos" significa que es algo que haces, como un gusto o un habito que haces regularmente, no necesariamente lo que haces ahora mismo.

@Proprietor If it is more than one neighbor it should be "neighbors" not "neighbor's"
