In general, a den is where a mammal (bear, fox, raccoon) lives and raises its young. It is a hole in the ground accessed by a short tunnel.
A nest is where a bird lives and raises its young. A nest is normally in a tree. Some mammals, like squirrels, also make a nest in a tree.
So, with a few exceptions, den means mammal and underground. Nest means bird and tree.
"The bear spent the whole winter hibernating in its den."
"I have a nest of bluebirds n the the tree outside of my bedroom window."
Sometimes, these words are used to describe where people (especially evil people) live or congregate. In this case, the words can be used interchangeably.
"That neighborhood is nothing but a den of thieves. You should avoid it"
"That neighborhood is nothing but a nest of thieves. You should avoid it"

Den can mean デン、書斎
“The mother fox retreated to her den.“
“I often think of this spot in the library as my own little den.”
Nest is like 巣(英語には蜘蛛と繋がってない)、ネスト
“The bird flew back to its nest.”
一番大きな違いは”nest”はただの名詞ではありません。 動詞でもあります。
“Owls often nest in barns.”
