One is in the present tense the other in the past. But even If I put both in the present, I would need some more context. "I am pleased about that" sounds perfectly natural, and "I am glad of it" not so much. They mean the same, if we were to use "I am glad of it", that I would not.
You could say
→ "I am glad of knowing that"
→ "At the time, I was glad of being there".
