Because in the old Batman film from the 80's, actor Jack Nicholson played the Joker, a character with a notoriously big, evil, grin. So if someone said "the biggest grin this side of Jack Nicholson's Joker" they are saying that someone has a huge grin like the character in the film.

No I normally do not say "this side of", I think it is more of an older expression that is normally not used anymore

It's definitely an older expression. Oftentimes people from the southern parts of the states are use these phrases in movies and stuff, but I'm not sure how accurate that is in real life.
