Is using "y usted?" in replying to "como estas?" or "how are you?" more formal than using "y tu?". For example I would use "y tu?" with a friend and "y usted?" with an older person

yes. "¿y usted?" is a pilite way to say "¿y tú?". both mean AND YOU?

Exactly, "usted" is more formal.
Usted is mainly used when talking to elders, people you don't know, or people that are in a higher rank than you like teachers or your boss. But lately its usage has been kind of limited to address only elders.
But "traditional, kind of old-fashioned" people like me feel weird when addressing teachers or unknown people with "tu" so i still keep "the rule" and only use "tu" with my friends or close family :p
