I know it's a strange question but I make this sentence interrogative.
Can I say "The more you learn, the less do you know?"? or just add "?"to the end?

Depends on what you're trying to ask. It can be, "Is it true that the more you learn, the less you know?"
Putting a question mark after the quotation isnt correct

You can ask "the more you learn, the less you know?" (with a "?" and with the inflection of asking a question)
Or you could add "so" beforehand: "So, the more you learn, the less you know?"
(To answer your question, I would just say it with a ? and say it like it is a question. You can also add something like "so" before it, or add "right?" onto the end - something like that)
