¿Cuál es la diferencia entre I am at my aunt's house. y I am in my aunt's house. ?Puedes dar oraciones como ejemplo.

I suppose there is no difference between them. Which one would you use?

@LoooooN: they're basically the same thing except some people might say 'at my aunt's house' when they have arrived, e.g. they are outside it

I think it's unusual to say "I'm "in" my aunt's house" unless there's a reason to specify "in" (as opposed to just outside or…on the roof?? lol). The assumption is that you will be inside. I can't really think of an example where it would be more appropriate to say "in" instead of "at" though I can imagine there will be one or two situations where it's more fitting. I guess if you were on the phone with someone and they asked (assuming you were at your house) "do you have the book, XXXXX?" You might say "oh I'm not sure! I'm at/in my aunt's house right now so I'll have to check my shelves later." (And even then, I prefer "at" and it is equally fine to use in that sentence so I would just generally use "at".)
