The sentence is not formed correctly. I think it sounds better when you say “ It wasn’t until he was threatened with losing his job, that the comments about John’s work ethic really hit home.”
What I understand from this sentence is that John did not have a good work ethic and he is complacent about his job. He became serious only when he was threatened about losing his job.
