Talking is used for conversations, when people are chatting to one another. Speaking is for more informative situations.

"I am talking on the phone"
"They are talking to their friend"
"I tried to talk him out of it" ( i tried to convince him not to do something)
"They are the talk if the town" (They are what everybody in the town is talking about)
"Can I talk to you?" (informal)

"Can I speak with you?" (more formal)
"Do you speak English?"
"Who is tonights speaker?" (who is going to be talking to the crowd tonight?)
